Teaching responsibilities will include an advanced undergraduate computational biology course, courses in the Professional Sciences master’s biotechnology curriculum, and other CMB offerings. The successful candidate will be expected to maintain active scholarship and participate in community/university service. Research involving undergraduate and/or Master’s students is expected.
PhD in biology, computational biology, bioinformatics, mathematics or related field and excellent communication skills are required. postdoctoral experience, experience in biotechnology, demonstrated success in teaching and research involving students are desired.

小分子的有机分子与大分子相互作用

小分子的有机分子与蛋白质的相互作用,是药物设计的基础

西药基本上是有机小分子(ligand,L)。我们吃的药一般都是作用在有机大分子蛋白质(protein,P)上, 形成符合结构(complex,C)。

L + P <=> C

人们用Kd(Disassociation Constant)

Kd=[L][P]/[C]

来表示大分子与蛋白质的亲和性。它的含义是当溶液中已有一半的蛋白质与小分子结合成符合结构时,即L的浓度与 C的浓度相等时,小分子的浓度.

Kd越大,两者就越容易分离,结合的时间也短。Kd越小,两者就越容易结合,而且不容易分开。人体中本来就有各种的小分子和各种的蛋白质相互作用,Kd一般的范围在nanomolar(nM)到micromolar (μM)。

如果一种药物能够作用在人们不希望它结合的蛋白质上,就会产生副作用,就成了毒药。所以人们研究的药物和期望蛋白质的Kd基本上是在0.1-10nM的非常强的范围,以尽量减少副作用。
